“My interest is in the first drug that we have a number of times,” Johnson said. “It’s important to me that we have a lot of that in the future.”
Johnson and his colleagues at the Institute for Medicinal Products of the American Medical Association were among the first scientists to work on an experimental compound to be approved for use in the US market. But they also developed a drug to treat a condition known as benign prostatic hyperplasia, or BPH.
In their investigation, Johnson and his colleagues reviewed pharmaceutical research documents and other documents produced by Pfizer. They looked at the documents for five years and found several key points. Some showed that the drug works in two ways — it blocks an enzyme that regulates hormone levels and it’s safe. In other, it does not cause an allergic reaction. The documents also showed that the drugs may increase the risk of developing erectile dysfunction, a condition that affects nearly 90 percent of men. Johnson and his colleagues used these results to develop a drug to treat BPH. That drug was approved in the US in 1998. Johnson said that the drug is the first of its kind to be approved to treat BPH, a condition that affects 80 percent of men over 40.
The drug was developed using a compound known as Tadalafil, which is similar to sildenafil (also sold under the brand name Cialis).
Tadalafil was first approved as an antidepressant in the early 1980s. It was approved by the FDA in 1998 as a treatment for pulmonary hypertension. It was first approved for use in the US in 2002 as a treatment for benign prostatic hyperplasia, or BPH.
The new drug was originally developed to treat erectile dysfunction in men by inhibiting an enzyme known as phosphodiesterase-5 (PDE5) in the body. In addition to Johnson’s work on the drug, the researchers involved in this new series will be conducting their own studies, which involve testing the effectiveness of the drug against a range of different types of BPH. Johnson said that his research was done to determine whether Tadalafil was effective in treating BPH. And he and his colleagues also planned to conduct experiments to test the drug’s effects on men with erectile dysfunction.
A total of 11,976 patients were screened in a multicenter, double blind, placebo-controlled study that followed more than 3,000 men suffering from erectile dysfunction or BPH. Those men received either placebo or a combination of tadalafil 20 mg and sildenafil 60 mg (sold as Revatio), which were then taken for one month.
Johnson said that his study was conducted in three phases of his work, starting with the early phase. This was a phase 1 trial of tadalafil in patients who had mild to moderate BPH. The first phase, which involved a single dose of 20 mg, followed by a two-week washout period. The second phase, which involved a placebo, was followed by a two-week washout period.
